The problem we're having here is trying to explain what you need to do, knowing you have no idea what's happening inside the engine. If you were closer, I could fix it in 5 minutes. But I can't explain what you have to do, sorry...Do you know that, with a 50 character limit, it's

Don't take it to Toyota, they will fleece you.
It's really not that hard, don't worry, if its not running you won't damage anything buggering around with just a spanner on the crank.
If the marks are aligned it MUST turn over, are you sure its not just compression? It'll tighten up, but then carry on, but you can tell its not actually mechanically touching as you'd hear a clonk if it does.
Take it back to the start and line the marks up, it doesn't matter where you start but you won't be able to line one mark up then the other if you turn it without the belt connected, you can do the crank till the pistons touch a valve, then turn the cam a bit more, then turn the crank a bit more till you have both marks line up again, you may have to try this a few times with the cam in different start positions.
You can leave a spanner on the cam and move it clockwise very slightly to get the belt on easier, move the cam back till the belt is tight and both marks still line up.
Leave the tensioner itself off till you have the belt on, you can hold the tensioner pully up with a screwdriver handle between it and the fan belt pully to stop the belt slipping off the cam and crank pulley as it will if the belt is left baggy round the tensioner pulley.
Once you've released the tensioner, using the crank pulley move it forward a bit, and watch that the cam and pump pulley move at the same time. If you want you can move it backwards back past the marks then move it back in the right direction and see if the marks line up properly, if so give it a full turn (x2 turns on crank) and it should all line up again.4x4toys.co.uk - Keeping you on and off the road...

As Tony says you should be able to distinguish between locking up from compression or something is making contact with pistons and valves. You can actually feel the metal to metal contact when it happens.
